Introduction
Following periodic acid oxidation, mucopolysaccharides in tissue basement membranes or in the cell walls of microorganisms such as fungi and Pneumocystis carinii are oxidized to expose aldehyde groups. These aldehyde groups reduce methenamine silver to black metallic silver. Sodium thiosulfate fixes the developed silver deposits and removes any unreacted silver ions. Light green is used as a background counterstain to enhance staining contrast.
