Introduction

Hematoxylin and Eosin Staining Solution (H&E) is mainly used to demonstrate the normal components and general pathological morphology of various tissues, allowing comprehensive histological examination. Hematoxylin and eosin staining is one of the most fundamental and indispensable staining methods in biology, histology, pathology, cytology, and related disciplines. It is widely used in pathological diagnosis, teaching, and scientific research and is of significant practical value.

Cell nuclei are mainly composed of acidic substances and therefore have a strong affinity for the basic dye hematoxylin. In contrast, the cytoplasm contains basic substances and has a greater affinity for the acidic dye eosin. After cells or tissue sections are stained with hematoxylin and eosin staining solution, the nuclei appear a distinct blue-purple color, while the cytoplasm, muscle fibers, collagen fibers, and other tissue components are stained varying shades of red. Red blood cells appear orange-red.
